This is a working overview of quality control, written for readers who want more than a one-paragraph summary but less than a textbook.

Routine quality control monitors retention time shifts, baseline noise, system pressure, and peak shape. Trends can reveal column aging, mobile phase preparation errors, detector drift, or sample degradation. Corrective actions may include replacing the column, preparing fresh mobile phase, or recalibrating the detector. Stability testing often uses HPLC to measure parent compound loss and degradation product formation. Open questions remain about how accelerated stability results extrapolate to long-term storage under varied conditions.
Quality control for HPLC testing combines scheduled checks, documented procedures, and review of results. Before sample analysis, system suitability testing confirms that the instrument, column, and method meet predefined criteria. Common criteria include resolution between critical peaks, retention time precision, peak tailing, and theoretical plate count. Failure triggers investigation before results are reported. Records link raw data, calculations, instrument logs, and analyst identity to each batch, supporting audits and repeat analysis.
Laboratories apply HPLC testing across pharmaceutical, food, environmental, and industrial chemistry. The method can measure active ingredients, impurities, additives, preservatives, and degradation products. Sample preparation often includes dilution, filtration, and sometimes extraction or derivatization. The choice of column, mobile phase, pH, temperature, and detector depends on the analytes and matrix. Results are compared with reference standards to assign identity and concentration. Method suitability is judged by resolution, precision, and accuracy.
HPLC testing is not a single fixed procedure; it is a family of separation modes. Reversed-phase, normal-phase, ion-exchange, size-exclusion, and affinity chromatography each suit different analyte properties. Reversed-phase methods dominate because they handle many neutral and moderately polar compounds. Detection can be optical, electrochemical, or mass spectrometric, and the detector dictates what information is available. Coupling with mass spectrometry increases selectivity and enables identification when standards are unavailable. The technique cannot separate every mixture without adjustment.
HPLC testing is an analytical technique used to separate, identify, and quantify components in a liquid sample. It relies on a pressurized mobile phase that carries the sample through a column packed with stationary phase. Different compounds travel at different rates because of interactions with the stationary and mobile phases. The resulting signal versus time is a chromatogram. Peak position indicates identity under specified conditions, while peak area or height relates to amount.
| Property | Value | Notes |
|---|---|---|
| Retention time RSD | ≤1% for five replicate injections | Typical criterion; method-specific limits apply. |
| Resolution | ≥1.5 between critical pair | Baseline separation is generally desired. |
| Tailing factor | ≤2.0 | Measures peak symmetry. |
| Theoretical plates | ≥2000 per column | Method-dependent; higher values indicate greater efficiency. |
| Peak area RSD | ≤2% for replicate injections | Reflects autosampler and detector precision. |
Data handling and documentation are central to HPLC quality control. Electronic systems should have audit trails that record changes to methods, sequences, and results. Integration parameters, such as peak baseline and threshold, can affect reported areas and must be defined in advance. Out-of-specification results trigger a structured investigation that may include reanalysis, instrument checks, and review of sample preparation. Regulatory inspections often examine raw data, audit trails, and training records to verify that reported results are traceable and reliable.
Method validation establishes that an HPLC procedure is suitable for its intended use. Key parameters include accuracy, precision, specificity, linearity, range, limit of detection, limit of quantitation, and robustness. Accuracy measures agreement with a true or accepted value, while precision describes repeatability and intermediate precision. Specificity confirms that the method measures the analyte without interference from impurities, degradants, or excipients. Validation is documented in a protocol and report, and acceptance criteria are set before experiments begin. Regulatory guidance varies by region, but the general principles are widely harmonized.
=== Seizures === For drug-resistant focal epilepsy, pregabalin is useful as an add-on therapy to other treatments. Its use alone is less effective than some other seizure medications. It is unclear how it compares to gabapentin for this use.
=== Mechanism of action === As an adenosine nucleoside triphosphate analog (GS-443902), the active metabolite of remdesivir interferes with the action of viral RNA-dependent RNA polymerase and evades proofreading by viral exoribonuclease (ExoN), causing a decrease in viral RNA production. In some viruses, such as the respiratory syncytial virus, it causes the RNA-dependent RNA polymerases to pause, but its predominant effect (as in Ebola) is to induce an irreversible chain termination. Unlike with many other chain terminators, this is not mediated by preventing addition of the immediately subsequent nucleotide, but is instead delayed, occurring after five additional bases have been added to the growing RNA chain. For the RNA-dependent RNA polymerases of MERS-CoV, SARS-CoV-1, and SARS-CoV-2, arrest of RNA synthesis occurs after incorporation of three additional nucleotides. Hence, remdesivir is classified as a direct-acting antiviral agent that works as a delayed chain terminator.
Two registrational trials have evaluated vaccine candidates in active immunization strategies against S. aureus infection. In a phase II trial, a bivalent vaccine of capsular proteins 5 & 8 was tested in 1804 hemodialysis patients with a primary fistula or synthetic graft vascular access. After 40 weeks following vaccination a protective effect was seen against S. aureus bacteremia, but not at 54 weeks following vaccination. Based on these results, a second trial was conducted which failed to show efficacy. Merck tested V710, a vaccine targeting IsdB, in a blinded randomized trial in patients undergoing median sternotomy. The trial was terminated after a higher rate of multiorgan system failure–related deaths was found in the V710 recipients. Vaccine recipients who developed S. aureus infection were five times more likely to die than control recipients who developed S. aureus infection. Investigators have suggested that a multiple-antigen vaccine would be more effective, but a lack of biomarkers defining human protective immunity keep these proposals in the logical, but strictly hypothetical arena.
The brown recluse (Loxosceles reclusa, Sicariidae, formerly placed in a family "Loxoscelidae") is a recluse spider with necrotic venom. Similar to those of other recluse spiders, their bites sometimes require medical attention. The brown recluse is one of two spiders in North America with dangerous venom, the other being the black widow. Brown recluse spiders are usually between 6 and 20 millimetres (0.24 and 0.79 in), but may grow larger. While typically light to medium brown, they range in color from whitish to dark brown or blackish gray. The cephalothorax and abdomen are not necessarily the same color. These spiders usually have markings on the dorsal side of their cephalothorax, with a black line coming from it that looks like a violin with the neck of the violin pointing to the rear of the spider, resulting in the nicknames fiddleback spider, brown fiddler, or violin spider.

=== Americas === Trump's second presidency has been characterized by a renewed American interference in the internal affairs of Latin American nations. These efforts have included both threats and promises made to influence the outcomes of elections in Honduras, Chile and Argentina, as well as tariff threats against Brazil and Mexico and military action against Venezuela.
Lattice models (e.g. BIO-LGCA models) Models similar to Dissipative particle dynamics that solve Newton's equation of motion with dissipative and random forces Models where cells depict Voronoi regions and an effective potential (based on Voronoi graphs) for the tissue is used Continuum models, e.g. with the use of a phase field Kinetic theories similar to the Boltzmann equation These mathematical models give some insight in complex phenomena like cancer, wound healing and ectoplasms.
The germ contains oil that is exposed by grinding, thus whole-grain cornmeal and grits turn rancid quickly at room temperature and should be refrigerated. Whole-grain cornmeal and grits require extended cooking times as seen in the following cooking directions for whole-grain grits: "Place the grits in a pan and cover them with water. Allow the grits to settle a full minute, tilt the pan, and skim off and discard the chaff and hulls with a fine tea strainer. Cook the grits for 50 minutes if the grits were soaked overnight or else 90 minutes if not." The milling of corn removes the aleurone and germ layers, removing much of the (already low) amounts of bioavailable niacin and tryptophan found within. The milling and degerming of corn in the preparation of cornmeal became feasible with the development of the Beall degerminator, which was originally patented in 1901 and was used to separate the grit from the germ in corn processing. Casimir Funk, who helped elucidate the role of thiamin in the etiology of beriberi, was an early investigator of the problem of pellagra. Funk suggested that a change in the method of milling corn was responsible for the outbreak of pellagra, but no attention was paid to his article on this subject.

=== Yeast === The specific yeasts and the environments in which they act determine the resultant organoleptic combinations. The role of yeast is, through many enzymatic processes, to turn sugars and carbohydrates into alcohol. There are two steps, first in aerobic conditions, yeast is doubled in colony size every four hours. This process goes on for 24–48 hours. Next, yeast turns acetaldehyde into ethyl alcohol, which is known as one of the organoleptic compounds produced in fermentation. The two main categories of yeast used in tequila are commercial brewers yeast and yeast that comes from precultivated existing yeast that has been preserved. The use of either type of yeast can result in different end products of tequila. Traditional production uses so-called "open fermentation", relying on yeasts from the surrounding environment. A 2023 article analyzes the diversity of yeasts found in these uncontrolled conditions.

BSA can be calculated on the basis of weight and height. Twenty-four-hour urine collection to assess creatinine clearance is no longer widely performed, due to difficulty in assuring complete specimen collection. To assess the adequacy of a complete collection, one always calculates the amount of creatinine excreted over a 24-hour period. This amount varies with muscle mass and is higher in young people/old, and in men/women. An unexpectedly low or high 24-hour creatinine excretion rate voids the test. Nevertheless, in cases where estimates of creatinine clearance from serum creatinine are unreliable, creatinine clearance remains a useful test. These cases include "estimation of GFR in individuals with variation in dietary intake (vegetarian diet, creatine supplements) or muscle mass (amputation, malnutrition, muscle wasting), since these factors are not specifically taken into account in prediction equations." A number of formulae have been devised to estimate GFR or Ccr values on the basis of serum creatinine levels. Where not otherwise stated serum creatinine is assumed to be stated in mg/dL, not μmol/L—divide by 88.4 to convert from μmol/Lto mg/dL.

Vitamin K is absorbed along with dietary fat from the small intestine and transported by chylomicrons in the circulation. Most of vitamin K1 is carried by triacylglycerol-rich lipoproteins (TRL) and rapidly cleared by the liver; only a small amount is released into the circulation and carried by LDL-C and HDL-C. MK-4 is carried by the same lipoproteins (TRL, LDL-C, and HDL-C) and cleared fast as well. The long-chain menaquinones are absorbed in the same way as vitamin K1 and MK-4 but are efficiently redistributed by the liver in predominantly LDL-C (VLDL-C). Since LDL-C has a long half-life in the circulation, these menaquinones can circulate for extended times resulting in higher bioavailability for extra-hepatic tissues as compared to vitamin K1 and MK-4. Accumulation of vitamin K in extra-hepatic tissues has direct relevance to vitamin K functions not related to hemostasis.
=== In the media === In 2007 the first website for Mendonça was created. In the same year, Mendonça was listed in the "Eight Portuguese Cases", of the newspaper Diário de Notícias, with cases of children that disappeared and remain unsolved. On Mendonça's 16th anniversary, Filomena Teixeira wrote a letter to her missing son. The letter was available on social media and in the Diário de Notícias. On Mendonça's 27th anniversary, a video was released on January 28, 2014 that became popular on social media. The actors Paulo Pires and Ana Padrão, among other people participated. In January 2020, Teixeira also wrote a second letter, this time for the 33rd birthday. An update to the PJ's website made it that Mendonça was taken out of the online missing people list for a few hours in August 2017.

System suitability is typically performed before each batch or according to the validated method and laboratory procedure. Some long runs include periodic checks during analysis. The required frequency depends on regulatory expectations and method performance.
Retention time drift can result from changes in mobile phase composition, column temperature, pump flow, or column age. A gradual shift often points to column degradation. A sudden shift may indicate a leak, mixing error, or incorrect mobile phase.
Retention time alone cannot confirm identity because different compounds may elute at similar times. Coupling HPLC with mass spectrometry or comparing against authenticated standards increases confidence. Confirmation usually requires orthogonal data.
It measures the presence and amount of one or more compounds in a liquid sample. Separation occurs in a column, and detection produces a signal proportional to concentration. Identification usually requires comparison with a known reference standard under the same conditions.
